Photomicrography

Photography through a microscope, recording subjects at magnifications far beyond macro.

Beyond about 10:1 a macro lens can no longer deliver useful resolution, and the microscope's objective takes over. Depth of field is measured in micrometres, so focus stacking is essentially mandatory, and illumination technique — brightfield, darkfield, polarised, phase contrast — determines what the image reveals as much as the optics do.

Examples in practice

A stack of 200 frames through a 20× objective renders a diatom sharp across its full depth.
